The Potosi mines (today Bolivia) in the Viceroyalty of Peru was the most productive silver mine for the Spanish Empire during the 16th century (which tracks with the time Drake was active in the Caribbean). However production was not as large during the remainder of the centuries of the Spanish Empire in the Viceroyalty of Peru.
Instead silver production rose in the Viceroyalty of New Spain (today Mexico), to become the main producer of silver for the Spanish Empire between the 17th and 19th centuries. So much so, Mexico City sent vast sums of silver to Spain to rebuild the Alcazar royal palace in Madrid, that burned down in the 1700s. It was in Mexico City that a lot of the piece of eight coins were minted to a standard. A lot of that minted silver was used to trade with China and Japan along the Manila-Acapulco galleon route (the Philippines were administered through Mexico City). The coins were also used in the British North American colonies.
The king of Spain gave a few Mexican cities their charters, creating the Camino Real (Royal Road), or informally the silver route which stretched from Mexico City to eventually New Mexico and Texas. Along the way, many silver mining cities were established, the most productive ones Zacatecas (eventually becoming the biggest silver producing region for the Spanish Empire), Guanajuato, San Luis Potosi, Durango. All along the silver route that transported all the extracted silver back to Mexico City, to be minted or be made into ingots, then shipped from the ports of Veracruz onto Cuba and Spain.
There were other silver cities further south not on this route, namely Taxco and Guadalajara.
It was between the viceroyalties of Peru (Bolivia) and New Spain (Mexico) together that it is thought that around 80% of the mined silver globally then was extracted from these regions between the 16th and 19th centuries.
